For MUAs and webmail, I don't see any alternative to manual testing, since 
it's all about bits of the user interface which are not standardized at 
all.

On the other hand, for tests of MTA, MSA, and MDA, those are about how 
well they implement the SMTP, submission, IMAP, and POP protocols.  In my 
tests those were entirely automated except for a few cases where the 
system I was testing was so broken that it wasn't worth adding code to the 
test scripts to work around it.
