I am thinking we should strike a balance on the two, because we could have a situation where someone who is new to twitter and has less followers but they end up with lots of tweets, this would result in the impact being minimal as opposed to those with more followers and more retweets to achieve the noise we want for the ICANN64.

I like one if Amrita's suggestions that the prize should go to the person who tweeted the most,  this way more tweets about ICANN64 is disseminated as I believe the whole point is to promote the program as much as possible, and getting people to tweet as many times a day will surely make ICANN64 a trending topic.
If we focus on most retweeted tweet basically the person with the most followers will always win. And most people will just be focusing on just tweeting just one tweet that is catchy.
Let's focus on tweeting as many times as possible every day from different time zones so any time you are in the twitterverse you should always see at least one tweet about the program.
As for the prize I agree with everybody.
